Abstract: 

Osteoarthritis (OA) is a degenerative joint disease usually seen in the elderly, which incidence increases with age. Its pathogenesis and underlying mechanism are still unclear. The disease severely affects the physical health and life quality of patients, thereby constituting a huge economic burden to family and society. Luteolin (LUT) is a natural flavonoid with multiple pharmacological properties. Many plants containing LUT have been applied in the treatment of several inflammation-related diseases due the relatively strong anti-inflammatory effects of LUT. The present study investigated the influence of LUT on cell apoptosis and inflammatory reactions in cartilage of OA guinea pigs, and its underlying mechanism. It was found that LUT effectively inhibited proliferation of OA cartilage cells, down-regulated the expressions of JNK and p38MAPK in cartilage cells of OA, and downregulated NO, TNF-α and IL-6. Thus, it alleviated inflammatory reactions, protected cartilage cells, and delayed cartilage degeneration.

Abstract: 

BACKGROUND: The roots of Korean red ginseng (Panax ginseng C.A.Mey.; KGC) have been used as an herbal supplement to enhance vital energy and immune capacity. Salvia plebeia R.Br. has been used to treat inflammatory diseases.PURPOSE: The aim of this study was to examine the anti-asthmatic effects of a mixture of Korean red ginseng and Salvia plebeia R.Br. (KGC3P), its component nepetin, and their modes of action in alleviating ovalbumin (OVA)-induced asthma in mice.METHOD: BALB/c mice were sensitized with OVA then subjected to intratracheal, intraperitoneal, and aerosol challenges. KGC3P and nepetin were administered orally for four weeks. Airway hyperresponsiveness (AHR), OVA-specific IgE levels, and Th2 cytokine- and gene expression levels in bronchoalveolar lavage fluid (BALF) and splenocytes were measured. Histological and immune cell subtype analyses were performed. PTEN and Akt phosphorylation levels were also evaluated.RESULTS: KGC3P reduced OVA-induced AHR, serum IgE levels, histological changes, and eosinophils infiltration but also the absolute number of immune cell subtypes including CD3/CD4, CD3/CD8, CD4/CD69, and Gr-1/CD11bin the lungs, BALF, and mesenteric lymph nodes (MLN). KGC3P also lowered the Th2 cytokines IL-4, IL-5, and IL-13 in the BALF and splenocytes and downregulated the IL-4, IL-13, IL-17, TNF-α, and MUC5AC genes in the lung. KGC3P upregulated the peroxisome proliferator-activated receptor (PPAR)γ gene but downregulated the p-Akt and p-PTEN phosphorylation. Similar results were obtained with nepetin treatment.CONCLUSION: KGC3P and nepetin are anti-asthmatic because they reduce various immune cells such as eosinophils and Th2 cell as well as Th2 cytokines. These mechanisms may be accompanied by the regulation of PPARγ expression and the PTEN pathway. Taken together, our results indicate that KGC3P and nepetin may potentially prevent and treat asthma.

OBJECTIVE: Korean red ginseng was reported to have many biological effects like the antioxidant and the anti-inflammatory activities. Oxidative stress and neuro-inflammation play major roles in the pathogenesis of Parkinson's disease (PD). The current study aimed to investigate the protective effects of ginseng on rotenone-induced PD in rats.METHODS: Rats were randomly allocated into 4 groups: normal rats, rotenone control, ginseng+rotenone and ginseng only treated rats. The severity of PD was evaluated through locomotor activity perceived in the open field test, histological examination and immunohistochemical detection of amyloid-β in brain tissues, in addition to the biochemical assessment of tyrosine hydroxylase activity in brain tissues. Moreover, the following parameters were investigated for studying the possible mechanisms of ginseng neuroprotective effect: nuclear factor-κβ (NF-κβ), tumor necrosis factor-alpha (TNF-α), caspase- 3, lipid peroxides and reduced glutathione (GSH).RESULTS: Ginseng exhibited potent neuroprotective effect that was reflected upon the histopathological examination, marked improvement in the locomotor activity and through its ability to suppress the amyloid-β deposition in the cortex and striatum along with significant increase in the tyrosine hydroxylase activity. Ginseng successfully inhibited the NF-κβ inflammatory pathway in brain tissues beside the inhibition of other oxidative stress and inflammatory mediators. Furthermore, it exhibited antiapoptotic effect via the inhibition of caspase-3 expression.CONCLUSION: Ginseng could be a promising treatment in PD. It can suppress dopaminergic neuron degeneration through variable mechanisms mainly via inhibition of NF-κβ pathway in addition to inhibition of oxidative stress and apoptosis.

Abstract: 

The aim of this study was to evaluate the safety and anti-obesity effects of the Korean red ginseng extract in dogs. To this end, we fed healthy beagles a Korean red ginseng diet and/or snack for 8 weeks. The dogs were submitted to a thorough physical examination, complete blood count, serum biochemistry analysis, analysis of adipose tissue activity, and body fat-content analysis by computed tomography (CT). At the end of the study period, the dogs that were fed the ginseng extract-diet/snack showed a significant decrease in body weight, body condition score and leptin levels relative to the baseline value. The CT findings revealed a decrease in body fat content in dogs fed the ginseng extract diet but not in those fed the ginseng-extract snack. The results of blood analysis did not show any meaningful changes in any of the dogs. All dogs tolerated the diet/snack well, and there were no adverse events. Our results suggest that the Korean red ginseng extract diet can potentially serve as an anti-obesity diet for reducing fat mass in dogs.

Effect of Korean Red Ginseng in individuals exposed to high stress levels: a 6-week, double-blind, randomized, placebo-controlled trial.

Abstract: 

Background: To investigate the neurobiological evidence supporting the adaptogenic effects of Korean Red Ginseng in reducing the harmful consequences of stress using a double-blind, placebo-controlled trial.Method: Sixty-three subjects with high stress levels were randomized to receive an orally administered, double-blind, 6-week treatment with Korean Red Ginseng (n = 32) or placebo (n = 31). All participants underwent a comprehensive psychological evaluation using Beck Depression Inventory and Stress Response Inventory, cognitive evaluation using the continuous performance test, biological evaluation by measuring blood levels of lipids, catecholamines, inflammation markers, and heart rate variability at baseline and after 6 weeks.Results: At baseline, both groups showed no significant differences in age, sex, years of education, Beck Depression Inventory, and Stress Response Inventory. After 6 weeks, triglyceride levels were significantly increased within the normal limit in theKorean Red Ginseng group ( = 4.11, = 0.048), and the epinephrine level was decreased in this group ( = 4,35, = 0.043). The triglyceride increase was significantly associated with epinephrine decrease ( = -0.087, = 0.041), suggesting that Korean Red Ginseng may stabilize the sympathetic nervous system. In addition, we detected a significant group by time effect in the visually controlled continuous performance test, suggesting positive effects of Korean Red Ginseng on cognition.Conclusion: Korean Red Ginseng might help to stabilize the sympathetic nervous system and improve cognition in individuals with high stress.

There are abundant data regarding the health-enhancing properties of whole ginseng extracts and single ginsenosides; however, no study to date has determined the receptors that mediate the effects of ginseng extracts. In this study, for the first time, we explored whether the antiinflammatory effects of Rg3-enriched red ginseng extract (Rg3-RGE) are mediated by retinoid X receptorα-peroxisome-proliferating receptor γ (RXRα-PPARγ) heterodimer nuclear receptors.Methods: Nitric oxide assay, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazoliumbromide assay, quantitative reverse transcription polymerase chain reaction, nuclear hormone receptor-binding assay, and molecular docking analyses were used for this study.Results: Rg3-RGE exerted antiinflammatory effects via nuclear receptor heterodimers between RXRα and PPARγ agonists and antagonists.Conclusion: These findings indicate that Rg3-RGE can be considered a potent antiinflammatory agent, and these effects are likely mediated by the nuclear receptor RXRα-PPARγ heterodimer.

Background: The ginsenoside Rg3, one of active components of red ginseng, has chemopreventive and anticancer potential. Cancer stem cells retain self-renewal properties which account for cancer recurrence and resistance to anticancer therapy. In our present study, we investigated whether the standardized Korean Red Ginseng extract (RGE) and Rg3 could modulate the manifestation of breast cancer stem cell-like features through regulation of self-renewal activity.Methods: The effects of RGE and Rg3 on the proportion of CD44/CD24cells, as representative characteristics of stem-like breast cancer cells, were determined by flow cytometry. The mammosphere formation assay was performed to assess self-renewal capacities of breast cancer cells. Aldehyde dehydrogenase activity of MCF-7 mammospheres was measured by the ALDEFLUOR assay. The expression levels of Sox-2, Bmi-1, and P-Akt and the nuclear localization of hypoxia inducible factor-1α in MCF-7 mammospheres were verified by immunoblot analysis.Results: Both RGE and Rg3 decreased the viability of breast cancer cells and significantly reduced the populations of CD44/CD24in MDA-MB-231 cells. RGE and Rg3 treatment attenuated the expression of Sox-2 and Bmi-1 by inhibiting the nuclear localization of hypoxia inducible factor-1α in MCF-7 mammospheres. Suppression of the manifestation of breast cancer stem cell-like properties by Rg3 was mediated through the blockade of Akt-mediated self-renewal signaling.Conclusion: This study suggests that Rg3 has a therapeutic potential targeting breast cancer stem cells.

There are studies on the relationship between osteoporosis and hearing and balance system. The goal of this study was to compare the proliferation and osteogenesis induction properties of mesenchymal stem cells derived from healthy and osteoporotic individuals to better understand the healing properties of Korean red ginseng (KRG) and Panax ginseng-Avena Sativa-Tribulus Terrestris mixture (PAT).MATERIALS AND METHODS: Osteoporotic and healthy MSCs were isolated successfully in culture conditions. The proliferation levels of cells treated with different doses of KRG and PAT were compared by Water-Soluble Tetrazolium-1 (WST-1) assay. Alkaline phosphatase (ALP) assay was performed by selecting the most effective KRG dose in proliferation.RESULTS: Morphology of isolated cells and the expression of cell surface antigens have been detected as similar. The WST-1 assay showed that KRG was effective on the proliferation of osteoporotic cells. The levels of ALP in osteoporotic cells treated with KRG is increased depending on the differentiation day compared to healthy cells.CONCLUSION: KRG triggered an increase in intracellular ALP levels of osteoporotic MSCs. It suggests that KRG on osteoporotic cells is influential in stimulating osteogenesis and may be useful in osteoporotic patients.

BACKGROUND AND AIMS: Cyclophosphamide (CPh) is a frequently used drug, in human and animals for its immunosuppressive and anticancer potential. However, it is metabolized by the liver yielding damaging toxicants (to the liver itself and other non-target vital organs) via oxidative stress, apoptosis induction and finally necrosis. Since there is no escaping of using such harmful medications, we focused on alleviating its side-effects. Panax ginseng Meyer is a potent candidate, and we still lack adequate information on its hepatoprotective role against cyclophosphamide-induced liver-damage.METHODS: Here, we used P. ginseng (Korean Red Ginseng) compared to vitamin-E (natural antioxidant) in combating CPh-induced liver damage. Forty-eight albino rats were divided into 6 groups, Control, Ginseng, Vitamin E, Cyclophosphamide (CPh), CPh + Ginseng or CPh + Vitamin-E. Blood samples were taken for biochemical analyses and liver samples were collected for histopathology, oxidative stress evaluation, and gene expression analyses.RESULTS: In CPh group, typical CPh-liver damage was evident (higher levels of AST, ALT, ALP; lower albumin and total proteins levels; lower liver tissue concentrations of SOD, GPX and CAT and higher MDA; injured liver histopathological picture; and finally increased TNF-α, IL-1β and Caspase3 and decreased BCL-2 genes expression). All these were abolished with either P. ginseng or vitamin-E administration. However, P. ginseng was overall superior to vitamin-E, especially in restoring blood biochemical findings and damaged histopathological picture.CONCLUSIONS: Therefore, P. ginseng is a potent hepatoprotector (vitamin-E to a lesser extent) and should be considered where liver damage is expected secondary to damaging medications; as cyclophosphamide.

BACKGROUND: Chemotherapy-induced neuropathic pain (CINP) is a serious side effect of chemotherapy. Korean Red Ginseng (KRG) is a popular herbal medicine in Asian countries. We examined the therapeutic potential of intrathecally administered KRG for CINP and clarified the mechanisms of action with regard to 5-hydroxytryptamine (5-HT)7 receptor at the spinal level.METHODS: CINP was evoked by intraperitoneal injection of cisplatin in male Sprague-Dawley rats. After examining the effects of intrathecally administered KRG on CINP, 5-HT receptor antagonist (dihydroergocristine [DHE]) was pretreated to determine the involvement of 5-HT receptor. In addition, intrathecal 5-HT7 receptor antagonist (SB269970) was administered to define the role of 5-HT7 receptor on the effect of KRG. 5-HT7 receptor mRNA expression levels and 5-HT concentrations were examined in the spinal cord.RESULTS: Intrathecally administered KRG produced a limited, but a dose-dependent, antiallodynic effect. Intrathecally administered DHE antagonized the antiallodynia caused by KRG. Furthermore, intrathecal SB269970 also reversed the effect of KRG. No changes in 5-HT7 receptor mRNA expression were seen in the dorsal horn of the spinal cord after cisplatin injection. After injecting cisplatin, 5-HT levels were decreased in the spinal cord, whereas those of 5-HT were increased by intrathecal KRG.CONCLUSIONS: Intrathecally administered KRG decreased CINP. In addition, spinal 5-HT7 receptors contributed to the antiallodynic effect of KRG.
